چکیده

Ovarian follicular fluid contains both stimulatory and inhibitory agents that influence the growth and maturation of oocyte. In the present study, an attempt was made to isolate and study the biological properties of ovarian follicular fluid peptide(s) in buffaloes. Bubaline ovarian follicular was made steroidand cell-free. A protein fraction was obtained by saturation (30-35% level) of the follicular fluid with ammonium sulfate. The protein fraction was purified with Sephadex-G 50 gel filtration chromatography and a single peak was obtained in the eluant volume, which was lyophilized. SDS-PAGE of the lyophilized fraction revealed a single band and the molecular weight of the peptide was 26.6 kDa. The peptide stimulated the cumulus cell expansion and in vitro maturation rate of oocytes in buffaloes in a dose dependent manner when it was incorporated at different dose levels (0, 10, 25, 50, 100 and 1,000 ng ml of maturation medium). The basic culture medium consisted of TCM 199 with Bovine serum albumin (0.3%). The in vitro maturation rates were comparable to those obtained with a positive control medium (TCM 199+20 ng EGF ml+steer serum (20%)). Further purification and biological assays may throw more light on the nature and functions of this peptide. (Asian-Aust. J. Anim. Sci. 2005. Vol 18, No. 11 : 1557-1563)

Effect of ovine follicular fluid peptide on follicle, oocyte, and somatic cell culture in buffalo (Bubalus bubalis)

Follicle and oocyte growth is under the control of systemic gonadotropins and various local intraovarian factors. The present study was undertaken to examine the efficacy of peptide(s) isolated from ovine follicular fluid on preantral and antral follicles, oocytes, cumulus and granulosa cells, and oviductal epithelial cell growth in vitro in a heterologous species (Bubalus bubalis). Follicular ...

P-87: Effects of Bovine Cumulus Cell Co-Cul tire on Post Fertilization Development of Buffalo (Bubalus bubalis) Oocyte

Background: There is a considerable numbers of buffalo (Bubalus bubalis) in Iran which have an important role in rural life. The reproductive performance of buffalo is lower than cow. Co-culture with somatice cells improved the maturation rate of most of the species. The aim of the present study was to evaluate the effect of bovine cumulus co-culture on post fertilization development of buffalo...

Influence of nitric oxide on in vitro growth, survival, steroidogenesis, and apoptosis of follicle stimulating hormone stimulated buffalo (Bubalus bubalis) preantral follicles

Effect of sodium nitroprusside (SNP), a nitric oxide (NO) donor, on in vitro survival, growth, steroidogenesis, and apoptosis of buffalo preantral follicles (PFs) was investigated. PFs (200~250 µm) were isolated by micro-dissection and cultured in 0 (control), 10(-3), 10(-5), 10(-7), and 10(-9) M SNP. To examine the reversible effect of SNP, PFs were cultured with 10(-5) M SNP + 1 mM N(ω)-nitro...

Ovarian Teratoma in a Water Buffalo (bubalus Bubalis)

The present short communication puts on record a case of ovarian teratoma in a water buffalo along with detailed description of its ovarian biometry, follicular fluid composition and functional assessment. The 2.0×2.3×2.9 cm teratoma was found in the right ovary of 2.4×2.6×4.2 cm size. The teratoma was firmly encapsulated with cartilaginous tissue and its cross section revealed tufts of hair ma...
